This symposium will bring together professionals from formulation and process development, engineering, quality, commercial manufacturing, academia, and regulatory authorities to discuss key topics, including formulation development, semi-continuous approaches, continuous coating, risk assessment and control strategies, regulatory considerations, and the latest advancements in continuous biotech production. Through dedicated presentations and case studies, participants will gain valuable insights and share expertise in these critical areas. During the event IMA Active and BASF, will present “Modernisation of tablet manufacturing: from powder formulation design to production scale with Continuous Direct Compression”.  Pharmaceutical stakeholders are undergoing a transition from traditional to modern manufacturing, for achieving higher efficiency, improving quality, reducing costs, and increasing flexibility. Direct compression remains the preferred process for advanced tablet manufacturing. This work describes a Quality-by-Design (QbD) approach to evaluate formulation influence and critical process parameters (CPPs) across batch or continuous blending technologies, combined with full-scale direct compression, from prime quality excipients to the final tablet product. Ready-to-use coprocessed excipients minimize the number of ingredients reducing manufacturing complexity and processing requirements. Critical quality attributes (CQAs) including blend uniformity (by means of NIR), tablet weight, hardness, thickness, and API content were measured while were varied using a design of experiment protocol. The study demonstrates that QbD principles and experimental design models provide a robust basis to define the process operating space supporting a knowledge-driven technology transfer toward Continuous Direct Compression at production scale.
